Our Sequencing Technologies Group is well-integrated across JGI departments to ensure a robust offering of sequencing capabilities and products to our users. This includes scaling the technologies and enabling state-of-the-art sample processing and sequencing capabilities.
The Genomic Technologies Group, which includes the Sequencing Technologies team, relies on collaborations with Science Programs and Operations teams to enable the JGI’s focus on expanding molecular capabilities and functional genomics, scaling capacity through end-to-end automation — and further advancing sequence applications, single cell genomics, biodesign, and metabolomics capabilities.
Overview of sequencing products
The JGI operates highly scaled short- and long-read sequencing pipelines. This includes advanced sample management (quality assurance/quality control [QA/QC]), large-scale liquid handling automation, and sequence analysis.
Portfolio
The JGI is dedicated to optimizing sequencing technologies, protocols, and workflows by enhancing capabilities available to our users, increasing throughput, and reducing costs. For instance, the implementation of higher-throughput automated sequencing library preparation methods on the Hamilton Vantage Liquid Handler platform has significantly increased sample prep capacity for Illumina DNA and RNAseq, while also reducing costs.
- Genome: DNA
- For genome assembly, structural variation, genetic diversity (MAGs), and resequencing
- Transcriptome: RNA
- For gene annotation, metagenome discovery, and gene expression
- Epigenome: DNA-protein interactions
- For methylation, genome structure, gene regulation, and transcription factor binding
The Sequencing Technologies Group applies cutting edge molecular capabilities and state-of-the-art next-generation sequencing to enable biological discovery by the JGI and the wider scientific community of JGI users. Adopting the latest sequencing technologies has allowed the JGI to increase sequencing output while significantly lowering costs.
- Illumina NovaSeq X Plus: the JGI’s primary sequence generator, for short-read, high throughput environmental samples and transcriptomics
- PacBio Revio: for long-read genome assembly and methylation detection
Staff process samples, perform quality checks, and prep libraries for sequencing. They apply Lean Six Sigma practices to process optimization and development approaches, testing various platforms to see what works best for applications. The team works hand in hand with the Project Management Office to help ensure the JGI’s genomic capabilities are ready and applicable to users.
